چکیده

use of mutants of Saccharomyces cerevisiae that, while maintaining the aerobic growth characteristics of the parent wild-type, show a resistance to oligomycin (Avner & Griffiths, 1970; Griffiths et al., 1972). This resistance appears to be located in the ATPsynthesizing or -hydrolysing enzyme (ATPaset) of the inner mitochondrial membrane. As a less complex material than entire mitochondrial membranes is desirable for investigation of the difference in oligomycin-sensitivity and its possible cause, we have purified and compared an ATPase from wild-type and mutant yeast mitochondria.

Heterologous expression, purification, and biochemistry of the oligomycin sensitivity conferring protein (OSCP) from yeast.

Yeast Saccharomyces cerevisiae oligomycin sensitivity conferring proteins (OSCP) have been expressed in Escherichia coli. Heterologous expression results in production of a protein that is identical to yeast mature OSCP, including the absence of the initiating methionine residue. Yeast OSCP expressed in E. coli has been purified to homogeneity and it is able to reconstitute oligomycin-sensitive...

The largest mitochondrial translation product copurifying with the mitochondrial adenosine triphosphatase of Saccharomyces cerevisiae is not a subunit of the enzyme complex.

Mitochondrial adenosine triphosphatase isolated from a double mutant of Saccharomyces cerevisiae lacking cytochrome b apoprotein and subunit II of cytochrome oxidase does not contain the mitochondrial translation product (approximate molecular weight, 32,000) previously suggested to be a subunit of the enzyme complex.

Purification and properties of H+-translocating, Mg2+-adenosine triphosphatase from vacuolar membranes of Saccharomyces cerevisiae.

H+-translocating, Mg2+-ATPase was solubilized from vacuolar membranes of Saccharomyces cerevisiae with the zwitterionic detergent N-tetradecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate and purified by glycerol density gradient centrifugation. Partially purified vacuolar membrane H+-ATPase, which had a specific activity of 18 units/mg of protein, was separated almost completely from acid phospha...

The effect of changed sterol composition on the kinetics of mitochondrial membrane enzymes in the yeast Saccharomyces cerevisiae [proceedings].

The effect of sterols on the kinetics of membrane-bound enzymes was examined. Two strains of the yeast Saccharomyces cerevisiae were used for this purpose: the wild-type haploid strain, S288C, and the lipid auxotroph mutant Ole-3 (Resnick & Mortimer, 1966). The primary lesion in the Ole-3 mutant is the loss of 8-aminolaevulinate synthetase (EC 2.3.1.37) (Woods et al., 1975). Consequently, the m...

Cisplatin cytotoxicity is dependent on mitochondrial respiration in Saccharomyces cerevisiae

Objective(s): To understand the role of mitochondrial respiration in cisplatin sensitivity, we have employed wild-type and mitochondrial DNA depleted Rho0 yeast cells. Materials and Methods: Wild type and Rho0 yeast cultured in fermentable and non-fermentable sugar containing media, were studied for their sensitivity against cisplatin by monitoring growth curves, oxygen consumption, pH changes ...
